Intensive crash-course from in silico hints to hard wet-lab mined data recovered using following methods:
- Plasmid preparation (PCR, molecularcloning, sequencing, restriction digest)
- Prokaryotic and eukaryotictransformation (heat shock, electroporation, nucleofection)
- Transformants assessment (colony PCR,qRT-PCR, cellular cloning)
- Phenotype testing (Western blotting,protein protection assay, immunofluorescent confocal microscopy, live imaging, superresolution (SIM) microscopy)
- CRISPR/Cas9 based gene knock out
